AK057037 Expression in EFTs

Ewing’s sarcoma is the second most common primary malignant bone tumor in adolescents and children. Members of Ewing’s sarcoma family of tumors (EFT) harbor a common translocation (t11;22)(q24;q12). This results in the expression of EWS-FLI1, which encodes a chimeric oncoprotein believed to be the driving force behind tumorigenesis. Many EWS-FLI1 splice variants have been described, and additional translocation variants partner EWS with other members of the ETS family of transcription factors. No single biomarker exists to reliably distinguish EFT from other small, round, blue-cell tumors. Microarray gene expression profiling done by researchers at Childrens Hospital Los Angeles (CHLA) has identified an EFT-specific non-coding transcript (ncRNA) called AK057037 with important regulatory and tumorigenic functions. The CHLA study revealed that the AK057037 transcript is highly expressed only in EFT when compared to rhabdomyosarcoma, neuroblastoma, osteosarcoma, Wilms’ tumor, glioblastoma, and oligodendroglioma. In addition, their research showed strong expression solely in EFT compared to a panel of 11 normal tissues.

 

Assay Features

 

The SureDx™ EFT Diagnostic Kit is a quantitative real-time polymerase chain reaction (PCR)-based assay that uses primers and hydrolysis probes for quantitative detection of the AK057037 non-coding RNA transcript in total RNA isolated in tumor tissues.  It contains specially formulated blend of enzymes and buffers that enable first strand cDNA synthesis and quantitative detection of the AK057037 transcript in a single tube, in one step.  Some of its features include:

Equipment and Materials


The SureDx™ EFT Diagnostic Kit requires a real-time PCR instrument capable of detecting FAM and VIC fluorescent probes. This kit includes reagents required for the reverse PCR amplification/detection, as well as validated controls. Columns and reagents for RNA isolation are not included.

This test includes reagents required for the reverese transcription and qPCR amplification/detection, as well as validated reaction controls. Columns and reagents for RNA isolation are not included.
